If you are serious about accessibility, one thing you really need to address is colour contrast in your designs.
Guideline 2.2 of the
Web Content Accessibility Guidelines 1.0 requires that foreground and background colour combinations provide sufficient contrast when viewed by someone having colour deficits, or when viewed on a black and white screen. Two colours provide good colour visibility if the brightness difference and the colour difference between the two colours are greater than a set range. They suggest a
colour contrast algorithm, which is used by the guys at Juicy Studio to create this handy online
colour contast analyser.

When I get stuck on a design or need some inspiration I find this online
colour scheme generator tool invaluable. The flash application generates color schemes of several types. Every scheme is based on one (base) color, which is supplemented with additional colors making together the best optical impression. It even lets you view those schemes as you would if you suffered from various types of colour blindness.
